Amgen Leads Oncology Push With T-Cell Engagers as AI and Organoid Platforms Reshape Drug Development

Amgen detailed its T-cell engager and precision therapy strategy at Citi's 2026 summit. GT Biopharma, Onco-Innovations, Oncotelic, and Samsung Biologics also advanced AI and organoid platforms in drug development.

Amgen presented its oncology strategy at Citi's 2026 Virtual Oncology Leadership Summit, emphasizing T-cell engagers and precision therapies as its two main modality pillars, while simultaneously reporting strong quarterly revenue and full-year guidance. The company also noted that artificial intelligence is enhancing target identification, molecule design, and data analysis across its research and development efforts.

Amgen is prioritizing differentiated therapies and targeting hard-to-treat solid tumors as well as selected hematological malignancies. Its products include IMDELLTRA, a DLL3-targeted T-cell engager approved for second-line small cell lung cancer, with over 1,600 U.S. sites administering it and ongoing phase 3 trials exploring first-line metastatic treatment and a subcutaneous formulation. LUMAKRAS, a KRAS G12C inhibitor available for certain lung and colorectal cancers, is in phase 3 trials in combination with chemotherapy and other drugs. BLINCYTO, a CD19-targeted T-cell engager, is moving to earlier therapy lines and being explored in autoimmune diseases due to its B-cell depleting properties. Xaluritamig, a STEAP1-targeted therapy, is in phase 3 trials focusing on survival data in prostate cancer, with development including monotherapy and chemo-free regimens. The company said it remains cautious about new modalities such as antibody-drug conjugates while focusing on its strengths in biologics and chemistry, and it is collaborating with Disco Pharmaceuticals to explore new targets.

In its most recently reported quarter, Amgen delivered multi-billion dollar revenue, growing at a mid single to low double digit rate year on year, and remained profitable even as it increased research and development spending. The company's operating margin narrowed somewhat versus the prior year, and adjusted earnings per share came in close to or modestly above consensus. Amgen provided full year guidance ranges for revenue and adjusted EPS, expecting revenue to continue increasing versus the past year.

GT Biopharma, a clinical-stage immuno-oncology company, reported on June 1, 2026 that it has implemented AI-based tools across the discovery and engineering of its tumor-targeting NK cell engagers and multi-domain proteins, aiming to accelerate development while reducing cost. The company said the resulting efficiency gains are expected to push multiple new development candidates into pre-IND development in 2027, and that its AI initiatives support expansion of its pipeline beyond its current oncology focus over time. GT Biopharma's clinical programs include GTB-3650, a Phase 1 candidate for CD33-expressing blood cancers, and GTB-5550, a Phase 1 candidate for B7-H3-expressing solid tumors, with the first GTB-5550 patient dosed in May 2026, all built on its TriKE platform.

Shares of Onco-Innovations have surged 79% over the past 30 days to close at €0.70, as the Canadian biotech firm prepares for Phase 1 clinical trials. The company is validating manufacturing processes for its active pharmaceutical ingredient ONC010 to meet international quality standards for an investigational new drug application, and contract research organisation Nucro-Technics is running animal models of the company's PNKP inhibitor technology to collect pharmacokinetic and biodistribution data. Partner Avance Clinical is preparing to meet the U.S. Food and Drug Administration to align on study design, and Onco-Innovations targets the launch of a Phase 1 trial in Australia in 2026, where it can claim research rebates of up to 43.5% on development costs. The company's AI subsidiary Inka Health Corp is running a joint programme with AstraZeneca to predict how well cancer therapies will work in individual patients, and a separate agreement with GlaxoSmithKline focuses on mining real-world clinical data. Oncotelic Therapeutics, a clinical-stage biotech focused on oncology and AI-driven solutions, has completed the integration of approximately 28 million scientific abstracts into its PDAOAI platform, enabling real-time application within a robotics platform developed jointly with TechForce Robotics. The integration allows scientific knowledge to be directly embedded into automated workflows operating in regulated environments, and initial deployments are expected in the coming weeks. The company's chairman and chief executive said the combined platform is designed to improve operational efficiency and support compliance across pharmaceutical development and manufacturing.

Samsung Biologics presented its patient-derived organoid platform at the AACR Annual Meeting 2026, emphasizing its role in bridging the gap between preclinical findings and clinical outcomes. The platform integrates next-generation sequencing and clinical datasets, with collaboration with Samsung Medical Center providing access to patient treatment histories and response data. In a colorectal cancer study, 24 patient-derived organoids were tested with five anticancer agents, showing over 80 percent correlation between organoid-based IC50 analyses and conventional cell-based assays, as well as a meaningful correlation with patient treatment responses. The company also integrates its bispecific antibody platform, S-DUAL, with organoid-based validation systems and highlighted applications in immuno-oncology, including co-culture systems with immune cells such as PBMCs and NK cells. Samsung Biologics noted that the FDA Modernization Act 2.0 has eased requirements for animal testing and expanded the use of alternative models such as organoids.
