Allogene to present ALPHA3 interim futility analysis for cema-cel in first-line LBCL on April 13

Allogene Therapeutics said it will review the interim futility analysis from its pivotal, randomized Phase 2 ALPHA3 trial with cemacabtagene ansegedleucel (cema-cel) in first-line consolidation large B-cell lymphoma (LBCL) on Monday, April 13, 2026. The company said the live conference call and webcast will take place at 5:30 a.m. PT / 8:30 a.m. ET.

The April update is expected to report whether cema-cel can eradicate minimal residual disease, or MRD, in MRD-positive LBCL patients randomized to cema-cel versus observation. The analysis includes 12 patients per arm and will assess MRD status at day 45 post-randomization. The company also expects to provide some safety information with the data, including outpatient administration details.

Under ALPHA3, newly diagnosed LBCL patients receive standard frontline therapy outside the trial. Patients who achieve remission then receive an ultrasensitive MRD test using circulating tumor DNA, and patients who are MRD positive are randomized to either standard-of-care observation or treatment with cema-cel.

For the futility analysis, the company said it is targeting a 25% to 30% absolute difference in MRD clearance between arms, while noting the observation arm is expected to have a non-zero clearance rate of roughly 20% without intervention. The trial is being conducted with outpatient management, including lymphodepletion and infusion in outpatient infusion clinics for most patients.
