Phase 1 study of MP0317 shows tumor-localized CD40 activation in advanced solid tumors

A phase 1 study found MP0317, a FAP-targeting CD40 agonist, showed tumor-localized CD40 activation, a favorable safety profile, and immune remodeling in advanced solid tumors.

MP0317, a CD40 agonist that targets FAP, showed tumor-localized CD40 activation with a favorable safety profile and induced immune remodeling in the tumor microenvironment in a phase 1 dose-escalation study in patients with advanced solid tumors. The findings were presented as support for the need for further clinical evaluation in combination with complementary therapies.

The study was described as an open-label, nonrandomized, dose-escalation phase 1 study of tumor-localized CD40 agonism with MP0317, a FAP x CD40 DARPin, in patients with advanced solid tumors. The report said MP0317 reprograms the tumor microenvironment.

Additional cited research described preclinical data obtained with MP0317 and its mouse homolog, and another cited study said Fc-optimized CD40 agonistic antibody elicits tertiary lymphoid structure formation and systemic antitumor immunity in metastatic cancer.
