Encorafenib

Drug

Also known as: Braftovi, BRAFTOVI

Drug Profile

Encorafenib is an oral BRAF-targeted antineoplastic agent marketed as Braftovi, used in biomarker-selected melanoma, colorectal cancer, and non-small cell lung cancer settings, typically in combination regimens. It inhibits key kinases in the MAPK pathway and is prescription-only. The FDA first approved it in 2018.

Drug Class
Antineoplastic BRAF (RAF kinase) inhibitor
Approval Status
FDA approved in June 2018 for unresectable/metastatic BRAF-mutant melanoma combination therapy, with later approvals including metastatic NSCLC (2023) and accelerated mCRC combination expansion (2024)
Mechanism of Action
ATP-competitive RAF kinase inhibition with downstream ERK phosphorylation decrease and cell-cycle arrest
Brand Names
  • Braftovi
Indications
  • \Unresectable or metastatic melanoma with BRAF V600E/V600K mutation (with binimetinib)\
  • \Metastatic colorectal cancer with BRAF V600E mutation (with cetuximab, with/without mFOLFOX6 per approvals)\
  • \Metastatic non-small cell lung cancer with BRAF V600E mutation (with binimetinib)\
