Selinexor is marketed as XPOVIO, an oral nuclear export inhibitor used in specific relapsed/refractory multiple myeloma regimens and in relapsed/refractory diffuse large B-cell lymphoma after prior therapies. The DLBCL indication is under accelerated approval based on response rate pending confirmatory benefit. U.S. initial approval was in 2019.

Mechanism of Action
Reversibly inhibits exportin 1 (XPO1), causing nuclear retention of tumor suppressor proteins, reduced oncoproteins, cell-cycle arrest, and apoptosis.
