Drug Profile

JSKN003 is an investigational anti-HER2 biparatopic/bispecific antibody-drug conjugate with a topoisomerase I payload, being developed for advanced solid tumors. It has shown promising safety and antitumor activity in studies in China and Australia, and U.S. FDA has cleared a Phase II study in platinum-resistant ovarian-related cancers. Multiple Phase III studies are ongoing in China.

Drug Class
Anti-HER2 bispecific/biparatopic antibody-drug conjugate (ADC)
Approval Status
Investigational candidate; has FDA IND-cleared Phase II studies and expedited designations, but no approved marketing status stated.
Mechanism of Action
Targets two HER2 epitopes, then after internalization releases a topoisomerase I inhibitor that stabilizes DNA-topoisomerase I cleavage complexes and causes tumor-cell apoptosis.
Indications
  • \Platinum-resistant recurrent epithelial ovarian cancer, primary peritoneal cancer, or fallopian tube cancer\
  • \Advanced HER2-expressing solid tumors including breast cancer and colorectal cancer (investigational)\
