Drug

Drug Profile

Crizotinib is an oral anti-cancer tyrosine kinase inhibitor marketed as Xalkori for ALK- or ROS1-positive non-small cell lung cancer and additional ALK-positive malignancies. It inhibits c-Met/HGFR, ALK, and ROS1 signaling pathways. It is prescription-only in the U.S. and EU.

Drug Class
Protein kinase inhibitor (ALK/ROS1/c-Met inhibitor)
Approval Status
FDA approved in August 2011 for selected late-stage ALK-positive NSCLC, with later FDA ROS1-positive NSCLC approval in March 2016
Mechanism of Action
Competitive kinase inhibition at ATP-binding pockets; inhibits ALK fusion activity and other target kinases
Brand Names
  • Xalkori
Indications
  • \Metastatic non-small cell lung cancer (ALK-positive or ROS1-positive contexts)\
  • \Relapsed/refractory systemic ALK-positive anaplastic large cell lymphoma\
  • \Unresectable/recurrent/refractory ALK-positive inflammatory myofibroblastic tumor\
